Trehalose (α-D-glucopyranosyl-1,1-α-D-glucopyranoside) is a stable and nonreducing disaccharide; can be used as sweetener, stabilizer, humectant; has many applications in the food, pharmaceutical, cosmetic industries. production from maltose catalyzed by trehalose synthase (TreS) simple economically feasible for industrial-scale application. Reducing cost enhancing efficiency of TreS synthesis conversion to critical production. In this study, homologous was constitutively overexpressed Corynebacterium glutamicum ATCC13032 removing repressor gene lacIq fragment plasmid, expression could exempt inducer addition induction process. For cell permeabilization, Triton X-100 permeabilization agent, treatment time 3 h. system, permeabilized cells recombinant C. were biocatalysts, 300 g/L substrate, 173.7 produced within 12 h under 30 °C pH 7.0 conditions. addition, whole-cell biocatalysts showed promising reusability. This study provides safe, convenient, practical, low-cost pathway trehalose.
